Lansing fire officials arrived at the 1,600 block of Martin Street within two minutes of a fire, but were not able to save twin two-year-olds.
Firefighters say the fire was most evident in the rear of the house on the second floor, where the dead toddlers were found.
Officials say a baby sitter and two other children made it to safety.
Twenty-eight firefighters were called in to handle the fire and rescue, and the Lansing Fire Department is investigating for the cause of the blaze.
